ABSTRACT

Background Pneumoconiosis is the most serious occupational disease in China, and silicosis accounts for about half of it. Any intervention effect of physical exercise as the key and core of lung rehabilitation training on silicosis is still unclear. Objective To explore potential intervention effect of physical exercise on silicotic mice. Methods Forty SPF C57BL/6 male mice were randomly divided into four groups, 10 in each group, including a control group, a physical exercise group, a silicosis model group, and a silicosis model + physical exercise intervention group. Silicotic mouse model was established by using 50 μL SiO2 suspension (200 mg·mL−1). A treadmill was used to prepare mice receiving physical exercise at 0° inclination, 12.3 m·min−1, 60 min·d−1, 5 d·week−1 for 4 weeks. Pathological morphology of lung tissues was evaluated after hematoxylin-eosin (HE) staining; deposition of collagen in lung tissues was evaluated after Van Gieson (VG) staining; expression of p-protein kinase R-like endoplasmic reticulum kinase (PERK) was detected by immunofluorescence staining; expressions of cyclin dependent kinase inhibitors (p21) and p-p38 mitogen activated protein kinase (p38) were detected by immunohistochemistry. The protein expressions of endoplasmic reticulum stress signal factors [p-inositol-requiring enzyme-1α (p-IRE-1α), p-PERK, and p-eukaryotic initiation factor-2α (p-eIF-2α)], senescence signal factors (p-p53, p21, and p16), mitogen-activated protein kinase (MAPK) signal factors [p-p38, p-extracellular regulated protein kinases (p-ERK), and p-stress-activated protein kinase (p-JNK)] were detected by Western blotting. Results After designed acute SiO2 exposure, the images of micro computed tomography (CT) showed high density shadows in lung tissues of the silicotic mice and less shadows in lung tissues of the physical exercise intervention mice. After HE staining, the proportions of silicotic nodule area in lung tissues was (18.67±3.89) % in the silicosis model group, and significantly decreased to (8.78±1.05) % in the silicosis model + physical exercise intervention group (P<0.05). After VG staining, the proportion of collagen fiber area of lung tissues was (10.37±2.18) % in the silicosis model group, and significantly decreased to (4.35±0.89) % in the silicosis model + physical exercise intervention group (P<0.05). The results of immunofluorescence staining showed that in the silicosis model group, the expression of p-PERK increased at the location of silicotic nodules, while in the silicotic model + physical exercise intervention group, the expression of p-PERK decreased. The immunohistochemical staining results showed that the expression of p21 and p-p38 increased in the lung tissues of the silicosis model group; the expression of p21 and p-p38 decreased in the lung tissues of the silicosis model + physical exercise intervention group. The results of Western blotting showed that compared with the control group, the expression levels of p-IRE-1α (0.11±0.03), p-PERK (0.95±0.40), p-eIF-2α (3.53±0.91), p-p53 (1.78±0.07), p21 (1.98±0.10), p16 (1.26±0.17), p-p38 (0.41±0.09), p-ERK (0.42±0.05), and p-JNK (3.20±1.23) of the silicosis model group were all upregulated (P<0.05). Compared with the silicosis model group, the expression levels of p-IRE-1α (0.03±0.01), p-PERK (0.31±0.12), p-eIF-2α (0.30±0.06), p-p53 (0.76±0.08), p21 (0.18±0.11), p16 (0.70±0.24), p-p38 (0.03±0.00), p-ERK (0.19±0.03), and p-JNK (0.46±0.21) of the silicosis model + physical exercise intervention group were downregulated (P<0.05). Conclusion Physical exercise may alleviate pulmonary fibrosis in silicotic mice, and inhibit abnormal expressions of endoplasmic reticulum stress signal, MAPK signal, and senescent signal.

ABSTRACT

OBJECTIVE:To systematically evaluate the effectiveness and safety of Shenfu qiangxin pills combined with chemical medicine conventional therapy in the treatment of chronic heart failure ,and to provide evidence-based reference for clinical drug use. METHODS :Retrieved from CNKI ,Wanfang database ,VIP,Google Scholar ,PubMed,the Cochrane Library and Embase database ,RCTs about Shengfu qiangxin pills combined with chemical medicine conventional therapy (trial group ) versus chemical medicine conventional treatment (control group )were collected during the inception to May 12th,2020. After literature screening and data extraction ,the quality of the literatures was evaluated with risk bias assessment tool recommended by Cochrane 5.1.0 system evaluator manual. Meta-analysis and sensitivity analysis were performed by using Stata 14.0 software. RESULTS:A total of 7 RCTs were included ,involving 596 patients. Meta-analysis results showed that the total response rate of trial group was significantly higher than that of control group [OR =4.14,95%CI(2.15,7.97),P<0.000 01];the results of sub-group analysis according to the different criteria for determining the efficacy showed that the total response rates of trial group determined by Lee integral method and cardiac function grading method were sig nificantly higher than that of the control group (P<0.05). After treatment ,N-terminal pro-B-type natriuretic peptide (NT-proBNP) level of trial group was significantly lower than that of control group [OR =-1.33,95%CI(-1.55, qq.com -1.11),P<0.000 01]. Results of sub-group analysis accor- ding to cardiac failure type showed that NT-proBNP level of patients with chronic heart failure in trial group was lower than control group (P<0.001). The level of left ventricular ejection fraction (LVEF)in trial group after treatment [WMD =5.76,95%CI (5.05,6.47),P<0.000 01] was significantly higher than control group ;after treatment ,the level of B-type natriuretic peptide [SMD=-1.61,95%CI(-2.58,-0.54),P<0.000 01],left ventricular end-diastolic diameter (LVEDD)level [WMD = -6.06,95%CI(-6.84,-5.27),P<0.000 01],left ventricular end-systolic diameter level [WMD =-0.52,95%CI(-5.70,-4.33), P<0.000 01] were significantly lower than control group. There was no statistically significant difference in the incidence of ADR between 2 groups(P>0.05). Results of sensitivity analysis showed that when NT-proBNP ,LVEF level ,LVEDD level after treatment were used as indicators ,there was no significant difference in the analysis results after eliminating heterogeneity source , compared with before elimination. CONCLUSIONS :Shenfu qiangxin pills combined with chemical medicine conventional treatment has good efficacy and safety.

ABSTRACT

Objective@#To improve the understanding of rare anti-myelin-associated glycoprotein (MAG) positive IgM monoclonal gammopathy related peripheral neuropathy (IgM-PN) .@*Methods@#Eleven cases of IgM paraproteinemia and anti-MAG antibody positive neuropathy diagnosed since 2014 in Peking Medical Union College Hospital were summarized. The medical records including clinical manifestation, lab results, treatment and prognosis were analyzed.@*Results@#Among the 11 patients (8 male and 3 female) , the median onset age is 63 years old (range from 52 to 77 years old) . The peripheral neuropathy of 9 patients were characterized by distal onset of numbness, 6 patients suffered from muscle weakness. The nerve conduction velocity study indicated that all 11 patients had demyelinating peripheral nerve damage, which was sensory predominant and more severe in lower limbs, 6 of them had secondary axonal damage. Monoclonal IgM gammopathy was identified in all 11 patients, among which 6 were IgM κ, 2 IgG κ and IgM κ bi-clonal, 3 IgM λ. Three patients were diagnosed with Waldenström’s macroglobulinaemia. The anti-MAG-IgM antibody was positive in all 11 cases. After diagnosis, 9 patients received combination chemotherapy including rituximab or rituximab treatment alone. The monoclonal IgM level declined significantly in 7 patients. The neuropathy was stable or improved.@*Conclusions@#Anti-MAG antibody positive IgM-PN is a rare M protein related disease. In peripheral neuropathy with undetermined etiology, we suggest to screen M protein and anti-MAG antibody. Chemotherapy including rituximab or rituximab alone is recommended as first-line therapy.

ABSTRACT

OBJECTIVE: observe the changes of localization and expression of acetylated tubulin alpha( Ac-tubulin α) in fibroblasts of silicosis fibrosis. METHODS: i) A total of 22 autopsy case with coal workers' pneumoconiosis( CWP) was selected as CWP group( autopsy lesion lung tissue) and their adjacent normal lung tissues surrounding fibrotic regions were selected as the control group using typical sampling method. ii) Specific pathogens free healthy male rats were randomly divided into control group and 5 dust-exposed groups with 10 rats in each group. In dust-exposed group,the rats were exposed to 2 000 mg/m~3 of silica dust for 0,2,4,8,12 and 16 weeks respectively by dynamic inhalation. The rats in control group were given no treatment. iii) Primary cultured rat lung fibroblasts were induced by Ang Ⅱ at a concentration of 100 nmol/L at different time points( 0,5,15 and 30 min,1,3,6,12,24 and 48 hours). iv) The expression of Ac-tubulin α and α-smooth muscle actin( α-SMA) were detected by immunohistochemical staining. The expression of Ac-tubulinα/Vimentin and Ac-tubulin α/α-SMA were observed by immunofluorescence staining. The relative expression ofα-SMA and Ac-tubulin α protein was detected by Western blot. RESULTS: The positive expression of Ac-tubulin α in lung tissue of CWP group was down-regulated while that of α-SMA was up-regulated compared with the control group( P < 0. 01). Immunofluorescence results showed that Vimentin positive expression in the CWP group was significantly increased in the lesion fibrosis area,while Ac-tubulin α expression was absent. The co-expression of Ac-tubulin α and Vimentin was detected in normal lung tissue of control group. The animal experiment results showed that the expression of Ac-tubulin αin lung tissue of rats was down-regulated and that of α-SMA was up-regulated with the prolongation of dust exposure time( P < 0. 01). The Ang Ⅱ induced the differentiation of fibroblasts into myofibroblasts,and the expression of Ac-tubulin α in fibroblasts gradually decreased and the expression of α-SMA gradually increased with the prolongation of Ang Ⅱ induction time( P < 0. 05). CONCLUSION: The lack of expression of Ac-tubulin α may be involved in the occurrence and development of silicotic fibrosis.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the inhibition effect and mechanism of N-acetyl-seryl-aspartyl-lysyl-proline (Ac-SDKP)on myofibroblast differentiation via regulating acetylated tubulin α (Ac-Tub α)in vivo and in vitro.</p><p><b>METHODS</b>Silicotic model were made by SiO2 douched and divided into 6 groups as follows: control (4w, 8w)group, silicotic model (4w, 8w)group and post-or pre-treatment by Ac-SDKP group. Pulmonary fibroblasts were divided into 5 groups: (1) control; (2) Ang II; (3) Ang II+Ac-SDKP; (4) Ang II+Valsartan; (5) Ang II+TCS histone deacetylase (HDAC)6 20b. The localization of Ac-Tub α and α-smooth muscle actin (SMA) were observed by immunohistochemical (IHC) and immunofluorescence staining. The protein levels of Ac-Tub α, α-SMA, collagen type I (col I) and HDAC6 were measured by western blot.</p><p><b>RESULTS</b>In silicotic nodules and interstitial fibrosis area, positive expression of α-SMA, a classical marker of myofibroblast, was ob-served by IHC, accompanied with absence expression of Ac-Tub α. Furthermore, Ac-SDKP post-treatment could attenuate the levels of col I, α-SMA and HDAC6 to 48.39%, 52.63% and 70.18% compared with the silicotic 8w group respectively. And in Ac-SDKP pre-treatment group, compared with the silicotic 8w group, these protein levels were decreased to 32.26%, 64.91% and 54.39% respectively (P<0.05). The up-regulation of Ac-Tub α was found in Ac-SDKP post-and pre-treatment and increased to 3.00 and 2.90 folds compared with the silicotic 8w group. Compared with control group, the levels of α-SMA, HDAC6 and col I in Ang II group were up-regulated to 1.66, 3.56 and 4.00 folds accompanied with down-regulation of Ac-Tub by 44.44% (P<0.05). Pre-treatment with Valsartan, TCS HDAC6 20b or Ac-SDKP could inhibited all this changes induced by Ang II in vitro.</p><p><b>CONCLUSION</b>Ac-SDKP can inhibit the myofibroblast differentiation and collagen deposition via sup-press HDAC6 and up-regulate the expression of Ac-Tub α in vivo and in vitro.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the mechanism of iridoid from Valeriana jatamansi treating irritable bowel syndrome.</p><p><b>METHOD</b>Sixty male SD rats were equally divided into 6 groups (2 controls, 1 model and 3 treatment doses) with 10 rats per group. The test groups were administered with iridoid (24.92, 12.46, 6. 23 mg x kg(-1)) while the control groups were administered with fluoxetine (2.5 mg x kg(-1), positive control) or distilled water (negative control). The model was established by chronic stress and independent feeding. The influence of iridoid from V. jatamansi on 5-HT and 5-HIAA in colon, serum and hypothalamic were observed in all groups.</p><p><b>RESULT</b>In the model group, the content of 5-HT in colon and serum increased significantly, but the content of 5-HT in hypothalamic decreased significantly. The content of 5-HIAA and the value of 5-HT/5-HIAA had no significant change. In three iridoid-treated groups, the content of 5-HT in colon and serum decreased, but the content of 5-HT in hypothalamic increased. The content of 5-HIAA had no significant change. The value of 5-HT/5-HIAA in colon and serum reduced.</p><p><b>CONCLUSION</b>The mechanism of iridoid from V. jatamansi treating irritable bowel syndrome may be related to the regulation effect to the levels of 5-HT from Gastrointestinal to central nervous system.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effects of hydroxy safflor yellow A (HSYA) on the expression of bFGF protein and MMP-9 mRNA or protein of transplantation tumor of gastric adenocarcinoma cell line BGC-823 in nude mice.</p><p><b>METHOD</b>The BGC-823 cells were subcutaneously injected into the right anterior armpit of BALB/C nu/nu nude mice, and the animal model of transplantation tumor was established. The experimental groups were treated with HSYA at concentration of 0.056 and 0.028 g x L(-1) and cyclophosphamide at 2 g x L(-1), or with physiologic saline. The tumor inhibitory effect was observed, and the mRNA expression of MMP-9 of transplantation tumor was detected by real time-fluorescent quantitation PCR and the protein expression of MMP-9 and bFGF were detected by enzyme linked immunosorbent assay.</p><p><b>RESULT</b>The IR in the group with HSYA at the concentration of 0.028 g x L(-1) is higher than in the group with normal sodium. After treatment with HSYA, the mRNA expression of MMP-9 has significant difference at the concentration of 0.028 g x L(-1) as compared with physiologic saline-treated group (P < 0.05), but the protein expression of MMP-9 and bFGF is obviously less than that in the physiologic saline-treated group (P < 0.05).</p><p><b>CONCLUSION</b>The possible mechanism of HSYA in given concentration to antagonize tumor angiogenesis may be related with inhibiting the protein expression of MMP-9 and bFGF or the mRNA expression of MMP-9 in tumor tissue to reduce the degradation of blood vessel basilar membrane, and to restrain the migration of blood vessel and decrease the tumor vascularization.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effects of Hydroxy Safflor yellow A (HSYA) on the growth of blood vessel of transplantation tumor of gastric adenocarcinoma cell line BGC-823 in nude mice and its underlying mechanism of antagonizing tumor angiogenesis.</p><p><b>METHOD</b>The BGC-823 cells was subcutaneouly injected into the right anterior armpit of BALB/C nu/nu nude mice and established the animal model of transplantation tumor. Then nude mice were divided into 4 groups at random: model group, control group, high or low dosage of HSYA group. The model group were treated with normal sodium by intraperitoneal injection, HSYA groups were treated with HSYA at concentration of 0.056 g x L(-1) and 0.028 g x L(-1) by intraperitoneal injection, and in these groups each mouse was injected 2 times everyday with 0.2 mL by 4-6 hours interval. The control group were injected in enterocoelia 1 times every 2 days starting from the third day with cytoxan at 2 g x L(-1). 20 days later, the volume and weight of nude mice were observed. The pathological change of tumor tissue was observed under optical microscope. The mRNA expression of VEGF and bFGF of transplantation tumor were detected by real time quantitative PCR.</p><p><b>RESULT</b>The volume (607.42 +/- 252.96) mm3, weight (0.88 +/- 0.14) g of transplantation tumor, the mRNA expression level of VEGF 0.49 +/- 0.13 and bFGF 0.60 +/- 0.48 are reduced significantly after treatment with HSYA at the concentration of 0.028 g x L(-1) compared with physiologic saline-treated group (P < 0.05 or P < 0.01). The tumor pathological angiogenesis of HSYA group is also less obvious than the normal sodium-treated group.</p><p><b>CONCLUSION</b>HSYA in given concentration can inhibit the growth of BGC-823 transplantation tumor, and decreasing the mRNA expression of VEGF and bFGF, which suggests that inhibiting tumor angiogenesis may be one of the mechanisms of HSYA antagonizing tumor.</p>

ABSTRACT

<p><b>UNLABELLED</b>To study the effect of a Chinese herbal prescription on external calcium deposition to weight-bearing bone in simulated weightlessness rats.</p><p><b>METHOD</b>Twenty-one male Wistar rats were divided into 3 groups: control group, tail suspension group, tail suspension with Chinese medicine group which takes a Chinese herbal prescription extract (containing Radix Rehmanniae Preparata, Radix Acanthopanacis Bidentatae, Radix Astragali, Radix Angelicae Sinensis, Concha Ostreae prepared by acetic acid) by intragastric administration. After 1 week adaption, there start off 3 weeks simulated weightlessness by tail suspension. At the eleventh day of simulated weightlessness, every rat was given one equal dose of 41Ca tracer by intragastric administration. Right femurs were separated as experiment over, and the ratio of 41Ca to 40Ca (41Ca/40Ca) was measured by accelerator mass spectrometry (AMS), while total femur calcium was measured by inductively coupled plasma-atomic emission spectroscopy (ICP-AES). Femur 41Ca deposition amount (DA) and femur 41Ca deposition ratio (DR) were calculated.</p><p><b>RESULT</b>The results showed that compared with control group, 41Ca/40Ca decreased significantly (P < 0.001) in tail suspension group, while in tail suspension with Chinese medicine group, it significantly increased (P < 0.05). DA and DR were both decreased significantly (P < 0.001) in tail suspension group, but no significant change in tail suspension with Chinese medicine group as compared with control group. Compared with tail suspension group, DA and DR increased significantly (P < 0.001) in tail suspension with Chinese medicine group.</p><p><b>CONCLUSION</b>Simulated weightlessness by tail suspension can cause decreased deposition of external calcium to weight-bearing bone, and the Chinese herbal prescription in this trial is effective to prevent the decrease. Moreover, multiple mechanisms may contribute to weightlessness induced osteoporosis, besides calcium deposition disturbance.</p>

Objective To study the effects of Chinese medicine compound on bone density, biomechanics, histomorphometry of weightlessness rats simulated by tail suspension. Methods Fifty Wistar rats were randomly divided into 5 groups with 10 rats each group:control group, model group, and low dose, medium dose and high dose Chinese medicine compound treated suspension group, the experiment period was 21 days. BMD of femur and lumbar vertebrae were detected by dual energy X-ray absorptiometry. ABSTRACT

Objective To evaluate the semen quality of potential semen donors of college students and investigate their reproductive health status. Methods According to the screening procedure in the Basic Standard and the Technical Norms of Human Sperm Bank revised by Chinese Ministry of Health in 2003,all sperm samples of 355 potential donors were examined. Results 37 of 355 donors sperm(10.4%) were qualified to the normal standard revised by Chinese Ministry of Health;and the rest(89.6%) were abandoned,because the sperm numbers of 267 donors were not reached the identified semen standard of the human sperm bank in the first screen,26 donors(7.4%)were with abnormal health status,4 donors did not ejaculate and got samples,9 donors with normal semen quality did not take physical health examination,the normal shape percentage of 6 donors sperm was lower than 30% and the thawed revival percentage with(a+b) grade of 6 donors was lower than 60%.135 of 355 donors(38.0%) sperm quality were qualified to the male productive standard of WHO. Conclusion The results indicated that the sperm quality of undergraduate declines and the society should pay more attention to the reproductive health of college students.

ABSTRACT

Health food has been used for a long history in our country,and it has unique advantages and is an important part of national medical care delivery in China. Up to now,its use is not under administration of food safety law. This article analyzes the differences among health food,drug and common food,the advantages in theory,resource,safety and compatibility of health food,the laws and regulations of health food published by government,and the status of health food industry,in order to demonstrate the importance and urgency of health food being administrated under food safety law,for the purpose of obtaining the academic and legal consensus.

ABSTRACT

Objective: To provide foundation for clinical application through studying the drug action and mechanism of action of Lingdantongfengkang Capsule on anti-gout effects. Methods: Lingdantongfengkang Capsule was taken orally. The experimental methods were as follow: mouse ear swell induced by dimethylbenzene, mouse pain induced by injecting acidum aceticum in abdominal cavity, mouse hyperuricemia model induced by yeast. Results: Lingdantongfengkang Capsule could lessen mouse ear swell induced by dimethylbenzene, decreae mouse stretching frequencies induced by injecting acidum aceticum in abdominal cavity and antagonize animal serum uric acid,liveness of xanthinoxidase and urea nitrogen induced by yeast to mice.Conclusion: Lingdantongfengkang Capsule possesses the anti-gout effects.

ABSTRACT

Pa disease is often caused by the invasion of wind,cold,moist and heat.Its symptoms usually are pain,numbness,tumescence,muscle and vessel contracture,unfavourable ection and stretch,arthrentasis,function disuse and so on.Its clinical characteristics are chronicity,recurrent attacks and aggravating little by little.Its mutilation rate is high,and it hurts the patients physically and mentally.The clinical treating method should be based on di erentiation of syndrome and di erentiation of disease.Selecting Chinese materia medica reasonably will conduce to satisfactory curative e ect.

ABSTRACT

With the development of business of manned spaceflight in China,physiologic derangement in human body,a serious problem in spaceflight,is dangerous to astronauts,as well as the mission progression.With complex diseases,it is Traditional Chinese Medicine(TCM) by which we can look through the appearance of disease,and get the essence of change of yin and yang,qi and blood,and zang and fu organs in the body.In this paper,the pathogenesis of space physiologic derangement is studied by the sight of TCM theory of correspondence between human and universe,and discussed from the aspects of yin and yang,qi and xue,zang and fu.The author explored the TCM origin of physiologic derangement in space and the determination of treatment based on differentiation of symptoms and signs with TCM.This work is wished to be beneficial to the business of space exploration in our mother country.
